Yoon Yoo-sun began acting as a child. She made her screen debut around 1974–1975 in the film The Person I Must Meet (also listed as You Become a Star, Too). Early credits include The Flying Iljimae (1978), Colors of the Rainbow (1980), and various television appearances such as Tiger Teacher (1981).

A major turning point came in the late 1980s. Her performance in the KBS drama Land (1987–1989) earned her the Best New Actress award at the 25th Baeksang Arts Awards in 1989. In 1994 she starred in the film The Story of Two Women, winning Best New Actress at the 32nd Grand Bell Awards. The following year she received both the Excellence Award and Popularity Award at the KBS Drama Awards for Even If the Wind Blows.

Throughout the 2000s and 2010s she became a familiar face in supporting roles, often portraying mothers, aunts, or strong-willed women.
